Unit I:
Organization of genome - human, plant, animal and microbial genome. whole genome
assembly. Enzymes in Molecular Biology: Nucleases, Restriction endonucleases, DNA
Ligases, topoisomerases, gyrases, methylases, other modifying enzymes
Unit II:
Gene cloning vectors- Biology construction of Plasmids, lambda Bacteriophages,
Cosmids, Phasmids vectors. Binary and Co integrative vectors.
Unit III:
Basic Techniques in Gene Manipulation: Isolation and purification of Nucleic Acids -
Agarose Gel Electrophoresis; Blotting - Southern, Northern and Western blotting
Unit IV:
Gene cloning techniques: cloning strategies; Screening techniques of recombinants:
Direct methods and indirect methodsUnit V:
Characterization of transcriptome- micro arrays, DNA chips and SAGE technology. DNA
Profiling/DNA fingerprinting: DNA Markers in disease diagnosis and finger printing:
