Advanced Proficiency courses in Plant Biotechnology

Plant tissue culture is the in vitro technique of culturing a plant part viz. a protoplast, a cell, a tissue or an organ, in appropriate nutrient medium (static or liquid) aseptically. The science of plant tissue culture not only fulfills ones curiosity to understand plasticity and totipotency nature of plant cells, but being the most important part of plant biotechnology acts as a tool to produce millions of clonal plants in a small space within a short period of time and offers many diverse ways for plant improvement such as somaclonal variation, Agrobacterium mediated plant transformation etc.

• Media preparation
• In vitro culturing technique
• Primary Culture: - Seed, Embryo, Nodal, Internodal, Leaf , Storage organ, Nucellus, Anther, Shoot tip, Root tip, Suspension culture
• Sub culture: - Callus, Shoot induction, Root induction, Somatic embryo induction
• Somatic embryo viability test
• Production of Synthetic seeds
• Stress tolerance studies and biochemical estimations ( Salinity stress, Heavy metal stress, Drought stress)
• Protoplast isolation
• Agrobacterium mediated transformation
• Genomic DNA isolation from plant samples
• Plasmid DNA isolation from Agrobacterium
• Screening of transgenic plants using PCR
• RAPD analysis to analyze the chemically induced Somaclonal variation
• Secondary metabolite productiion, purification and confirmation through biochemical tests
